Sequencing run

Approx 40 Mins

Goal: to prime your flow cell with buffer, load your DNA library into the flow cell, and start a sequencing experiment in MinKNOW.

This step will take you through flow cell priming, sample loading and starting your sequencing experiment.

What you will need

  • 1.5 ml Eppendorf DNA LoBind tubes
  • Bovine Serum Albumin (BSA) (50 mg/ml) (e.g Invitrogenā„¢ UltraPureā„¢ BSA 50 mg/ml, cat# AM2616)
  • Flow Cell Flush (FCF)
  • Flow Cell Tether (FCT)
  • Lambda DNA library (Tube 1)
  • Library Beads (LIB)
  • Microfuge
  • P100 pipette and tips
  • P100 pipettes and tips
  • P1000 pipette and tips
  • P20 pipettes and tips
  • Sequencing Buffer (SB)
