Sequencing run

Approx 40 Mins

Goal: to prime your flow cell with buffer, load your DNA library into the flow cell, and start a sequencing experiment in MinKNOW.

This step will take you through flow cell priming, sample loading and starting your sequencing experiment.

What you will need

Bovine Serum Albumin (BSA) (50 mg/ml) (e.g Invitrogen™ UltraPure™ BSA 50 mg/ml, cat# AM2616)
Flow Cell Flush (FCF)
Flow Cell Tether (FCT)
Flow cell, MinION Mk1B, USB3 cable and compatible computer - set up in previous sections
Library Beads (LIB)
Microfuge
P100 pipette and tips
P100 pipette and tips
P1000 pipette and tips
P20 pipette and tips
Sequencing Buffer (SB)
Tube 1 - Lambda DNA library
Tube 3 - 1.5 ml Eppendorf DNA LoBind tube
