Library preparation

Goal: to use the Rapid Sequencing Kit to prepare a Lambda DNA library.
Library preparation using the Rapid Sequencing Kit is a simple, 10-minute procedure.
The protocol uses one 0.2 ml PCR tube and one 1.5 ml Eppendorf tube for the library prep, and two 1.5 ml Eppendorfs for flow cell priming and loading.
What you will need
- 0.2 ml thin-walled PCR tubes
- 1.5 ml Eppendorf DNA LoBind tubes
- Control Expansion (EXP-CTL001)
- Ice bucket with ice
- Microfuge
- Nuclease-free water (e.g. ThermoFisher, AM9937)
- P100 pipette and tips
- P1000 pipette and tips
- P2 pipette and tips
- P20 pipette and tips
- Rapid Sequencing Kit (SQK-RAD114)
- Thermal cycler or heat blocks
- Timer
- Vortex mixer
