Place sufficient absorbent material on the bench to take up approximately 4000 µl of flush waste.
-
Remove the flow cell light shield by sliding open the inlet port and gently lifting the light shield until it clicks off from the flow cell.
-
Place the flow cell onto the absorbent material at 45° angle. The flow cell should NOT be in the PromethION. Using a P1000, flush 1 ml of de-ionised water through the sample input port. Repeat 3-4 times, with fresh de-ionised water, taking care that all the liquid coming out of the waste reservoir is absorbed completely.
-
Once flushing is complete, close the sample input port cover and ensure that you have removed all the liquid from the reservoir by extracting the liquid using a pipette from the aspiration port. This will ensure that you leave sufficient liquid in the sensor chip area and neighbouring channels to submerge the sensor array during the return to Oxford Nanopore Technologies.
-
Dry any large liquid droplets around the flow cell ports.
If you have the old PromethION flow cell packaging, seal the aspiration ports with the sealing sticker included within the returns pack. Ensure a good seal to prevent leaking/evaporation of the water.
-
Dispose of the absorbent material as biological waste, as detailed in local guidelines.
-
Wipe the bench surface with a suitable disinfectant. Do not use any strong acids or alkalis.
