Prepare your priming mix
Make sure the contents of the Flow Cell Flush (FCF) and Flow Cell Tether (FCT) tubes are fully defrosted. Mix the tubes by pipetting up and down, and then spin down in a microfuge.
For optimal sequencing performance and improved output on R10.4.1 flow cells (FLO-MIN114), we recommend adding Bovine Serum Albumin (BSA) to the flow cell priming mix at a final concentration of 0.2 mg/ml. We do not recommend using recombinant BSA.
Transfer 30 µl of Flow Cell Tether (FCT) directly into one of the Flow Cell Flush (FCF) tubes.
Add 5 µl of Bovine Serum Albumin (BSA) at 50 mg/ml to the tube of Flow Cell Flush (FCF).
Mix the contents of the tube by flicking the tube with your finger and then spin down briefly in a microfuge.
The resulting buffer is now referred to as the Priming Mix (FCF tube). You will use this to flush out the storage buffer from the flow cell.
